       Trifluoroacetic acid (TFA, CF ∝ COOH) is a highly volatile perfluorinated organic acid with much stronger acidity than ordinary acetic acid. It is miscible with water and most organic solvents and is mainly used in organic synthesis, biomedicine, analytical chromatography, chemical engineering, and other fields. At the same time, it can be used to prepare downstream fluorine chemical intermediates such as trifluoroacetic anhydride and trifluoroacetic acid ester.
1、 Organic synthesis field
1. Strong acid catalyst: used for esterification, condensation, cyclization, rearrangement, alkylation, and isomerization reactions. Relying on strong proton acidity to catalyze reactions that are difficult to drive with ordinary acids; Low boiling point, can be removed by vacuum distillation at the end of the reaction, and is convenient for post-treatment.
2. Deprotection reagent: The most commonly used reagent for peptide synthesis, which cleaves and removes the Boc amino protecting group; It is also used to remove some protective groups from sugar and nucleoside molecules, completing molecular cleavage and releasing target products.
3. Reaction solvent: can dissolve many aromatic compounds, heterocycles, some polymers, and proteins that are difficult to dissolve in ordinary solvents; Used for fluorination, nitration, and halogenation reactions, changing the reaction pathway to achieve reaction selectivity that conventional solvents cannot achieve.
4. Synthesis of Fluorinated Intermediates: Used for the preparation of trifluoroacetic anhydride and ethyl trifluoroacetate, as raw materials for introducing trifluoromethyl structures, to produce fluorinated molecular frameworks for pharmaceuticals and pesticides.
2、 Biomedical and Peptide Synthesis
Solid phase peptide synthesis core reagent, used for the cleavage and cleavage of polypeptide chains on resins, while removing side chain protecting groups; It is a commonly used material for peptide raw materials, peptide intermediates laboratory, and small-scale production.
Attention: The residue of trifluoroacetic acid in the finished product needs to be strictly controlled. Excessive residue can cause toxicity and pH risks. Desalination and TFA removal processes must be carried out during production.
3、 Ion pair reagents for liquid chromatography analysis (HPLC)
When separating peptide and protein samples by reverse phase chromatography, low concentration trifluoroacetic acid (0.05-0.1%) is added as an ion pair modifier to the mobile phase.
Improve peak shape, prevent peak tailing and peak broadening;
Adjust the pH of the mobile phase;
Low UV absorption and minimal interference in low wavelength detection;
Having volatility, it is easy to remove reagents and obtain samples after preparing the liquid phase.
4、 Pesticide and dye chemicals
As a fluorine-containing intermediate raw material, synthesize herbicides and insecticides with trifluoromethyl structure; Trifluoromethyl can enhance the stability and biological activity of pesticide molecules; Also used for the synthesis of some high-performance fluorinated dyes.
5、 Other niche industrial uses
1. Used as a flotation reagent for non-ferrous metals in the mineral processing industry;
2. Material modification: Partial polymer and ion membrane modification additives;
3. In the field of scientific research, it can be used for protein dissolution and protein sequence analysis;
4. As a research direction for lithium battery electrolyte additives, it has not been widely industrialized.
